Several data processing stations with the following software : FlowJo, Diva, Kaluza, SpectroFlow.

1 clean room in L2 containment for cell sorting.

It is requested, for each publication that required the use of the device, that the department be listed in the acknowledgements.

Objective :

The initial objective was to open a high-performance flow cytometry center, able to provide multiparametric analysis and sorting. Integrated in the IRMB and being part of MRI, the laboratory participates in the work of regional laboratories. This laboratory, which does not have its own theme, is widely open to the regional scientific community: INSERM, CNRS, University of Sciences, Faculty of Pharmacy, IFREMER, IRD, CIRAD, INRA, ENSAM... In recent years, the technical evolution of the equipment has led many laboratories to analyze and purify cells by means of flow cytometry to better clarify their role or analyze their content (protein, DNA, RNA... ), whether human, animal, plant, bacterial, marine... The needs of laboratories are increasing with the evolution of technologies, many studies require the use of analyzers and high performance sorters.

Activity :

The flow cytometry service is a structure that operates as a common service.

What distinguishes FMC from other preparative techniques is that it offers the possibility to work simultaneously on several parameters. No other method, whether physico-chemical (gradient centrifugation, elutriation) or immunological (panning, columns, rosettes) offers this versatility. It is thus possible to analyze and sort populations on a multiparametric basis: two morphologies and up to 40 fluorescences on the latest generation of commercial instruments.
For the time being, the following topics (using flow cytometry analysis) are being addressed in response to the requests expressed:
+The study of the proliferation of various cell types (lineages, lymphocytes...) by DNA labelling.
+analysis of cell populations labelled with monoclonal antibodies (multiple labelling).
+ Quantification of membrane proteins by using calibrated standards (QIFIKIT).
+ study of apoptosis.
+study of protein expression according to the cell cycle.
+study of the DNA content of plants (lemon, coffee, rice, petunia...) by means of the dyes used for the marking of the DNA previously mentioned.
+Cellular sorting using the analysis techniques mentioned above.

Expertise :

The laboratory has participated in the testing of various devices from the 2 largest cytometer manufacturers, Becton Dickinson and Beckman Coulter.
